Notes:
1. Please provide templates in 0.2 ml flat-top PCR tubes, if possible. One such tube is Phenix cat. no. S9-MPC-200F. Write sample name on top with an extra fine-point Sharpie; use your initials, followed by sequential numbers, e.g., JS1, JS2, etc.
2. Investigator qualifies for "Project" rates by agreement with CIF upon written description of project, and with a minimum number of 10 plates (960 templates) per month. For projects involving sequencing only, templates are provided in 96-well plates as in 3. above. For projects with plasmid preps, investigator sends sealed, deep-well culture plates with bacterial cell pellets by overnight mail on blue ice. We use a liquid-handling robots (Beckman Multimek 96 and MWG Roboprep 2500) to automate plasmid preps. We can also plate, pick (we use a Genetix QPIX) and culture (Gene Machine Hi-Gro four-tower incubators) your library preparatory to extracting plasmid for sequencing or PCR reactions. Please inquire about this option.
3. Investigator provides templates preloaded in semi-skirted PCR plate, e.g., Phenix MPX-PCR96-HS. If plates are mailed (dry, only!), please seal tops; an appropriate seal is a thin adhesive film, e.g., Phenix LMT-THIN-EX. These rates will also be honored for 48 templates filling the first 6 columns of a PCR plate.
4. DNA sequencing reactions for this activity use 1/2 our normal amount of the most costly reagent, the Taq/fluorescent dideoxynucleotide mix; we pass on the savings to our clients with this reduced rate. The signal strength is reduced slightly from that in our full reaction, but read length is generally not affected or is reduced by only 20-30 bases.
